HepPar1 Antibody Differentiates Liver Tumor Types in Fine Needle Aspirations

NEW YORK (Reuters Health) Mar 15 – Fine needle aspiration (FNA) tissue samples from primary hepatocellular carcinoma and cholangiocarcinoma in the liver can be easily differentiated by using hepatocyte paraffin 1 (HepPar1) monoclonal antibody, according to research conducted in Dallas.
Dr. M. Hossein Saboorian and other pathologists at the University of Texas Southwestern Medical Center explain in the February 25th issue of Cancer Cytopathology that HepPar1 is a mouse monoclonal antibody that is highly specific for hepatocytes and only rarely reacts with bile-duct and nonparenchymal liver cells.

The researchers studied 75 hepatic tumor cell blocks obtained by fine needle aspiration, originally prepared between 1995 and 2000. The tissues were deparaffinized and hydrated, then incubated with HepPar1 (clone OCH1E5.2.10; Dako Company, Carpenteria, California) and goat antimouse secondary antibody. The sections were then stained with diaminobenzidine and counterstained with Harris hematoxylin.

The investigators report that all 50 hepatocellular carcinomas picked up the stain at moderate to strongly positive levels. Five cholangiocarcinoma sections were negative except for hepatocytes found between tumor cells, which were diffusely positive. Comparison with the original stained sections proved the hepatocytes to be benign.
